Required Qualifications: ALL REQUIRED QUALIFICATIONS MUST BE DOCUMENTED ON APPLICATION MATERIALS > Bachelor’s degree or equivalent in a scientific discipline, AALAC training as a Laboratory Animal Technologist and a mini mum of 5 years of experience working with a large, complex mouse colony. Necessary skills include mouse colony management, ability to design and implement preclinical studies to test novel compounds, drug administration (IM, IP, IV, oral gavage), blood collection, behavioral experiments, surgeries, health evaluations, animal imaging and necropsies including perfusion fixation. Molecular biology skills including DNA and RNA isolation, PCR, ELISA and mammalian cell culture are required.

Preferred Qualifications:
Special consideration will be given to applicants with experience in the isolation and culture of primary mammalian cells. The ideal candidate will have strong problem-solving skills and the ability to maintain responsibility for multiple projects simultaneously. An ability and willingness to train others in animal experimentation is desirable.

About the Job

Summary of Position:
This individual will take primary responsi bility for maintaining the laboratory mouse colony. This involves setting-up breedings to generate hundreds of different transgenic lines, weaning, earing, genotyping and organizing animals into experimental cohorts. The individual will be responsible for conducting genetic and pharmacologic studies aimed at impacting healthspan and lifespan. This includes measuring numerous functional, clinical and behavioral endpoints longitudinally in mice to determine their health status, as well as animal imaging. It also includes taking responsibility for organizing and performing large necropsies of mice to collect biological samples and distribute these samples amongst collaborators and our tissuebank. The candidate will be responsible for writing animal protocols, managing imports and exports, and examining the literature for state-of-the-art practices for measuring molecular and functional endpoints in mice. The individual must use our mouse cloud database to generate specimen labe ls and enter data. Preparation of tissue lysates, isolation of nucleic acids and protein, PCR, qRT-PCR, electrophoresis, ELISA, immunoblotting. Primary cell culture. Animal surgeries. Data analysis and contributing methods and figures to manuscripts and grant applications. Instruct others in all of the above practices and to create laboratory SOPs.

ESSENTIAL DUTIES:
70% Pre-clinical studies in mice (essential)
Design and implement genetic and pharmacologic studies aimed at testing hypotheses about fundamental mechanisms of aging. This includes management of a highly complex mouse colony with over 100 strains of mice, creating complex breeding schemes to obtain novel strains of experimental mice, quantitatively evaluating the health/frailty of live animals, designing and implementing serial behavioral and imaging studies, surgical techniques including stereotactic injections and parabiosis, organizing and leading mouse necropsies on large cohorts of an imals, collection and proper preservation of mouse tissues and bodily fluids, calculate dosing and the administration of drugs and special diets. Ordering all drugs, diets, specialized equipment and disposables needed to execute animal experiments.

10% Molecular biology (essential)
Mammalian cell culture, preparation of cell and tissue lysates, isolate DNA, RNA and protein, quantitate nucleic acids and protein levels, perform PCR genotyping, nucleic acid and protein electrophoresis, ELISAs.

10% Sample Management (essential)
Independently iteract with members of large programmatic grants, sponsored research agreements and collaborators to collate lists of biological samples needed. Supervise collection of all biological samples from pre-clinical mouse studies. Organize and execute distribution of biological samples to collaborators world-wide. Interface with our independent contractor to improve the workflow of sample management via the MouseCloud Da tabase.

10% Data Management (essential)
Supervise the entry of primary data into the MouseCloud Database. Analyse molecular biology and animal experimentation data. Provide methods and figures from this data for manuscripts and grants. Interface with our independent contractor to improve the workflow of data management and sharing via the MouseCloud Database.
